Sanofi said Kevzara – which it produces with partner Regeneron – had failed as a COVID-19 treatment after the latest set of trials in patients across the world showed adverse effects, and would be dropped for studies in this field.

Sanofi said the trials had led in some cases to COVID-19 pneumonia infections and even death in a few cases, Reuters reported.

The two companies do not anticipate conducting further clinical studies of it, Sanofi added.

Trials had shown that the drug did not help patients with less severe COVID-19, the companies had said in July.

“Although this trial did not yield the results we hoped for, we are proud of the work that was achieved by the team to further our understanding of the potential use of Kevzara for the treatment of COVID-19,” said Sanofi’s global head of research and development, John Reed.

“At Sanofi, we are committed to help combat the global COVID-19 pandemic, including developing vaccine candidates that can be manufactured at large scale,” he added.

Pharmaceutical companies have been racing to develop treatments against the COVID-19 pandemic that has claimed more than 849,000 lives and crippled economies.

Last month, CEO Paul Hudson said Sanofi’s confidence in its coronavirus vaccine candidates had increased over the summer as the drugmaker prepared to start clinical trials.

In July, Roche said its attempt to retool its rheumatoid arthritis drug Actemra/RoActemra to treat patients hospitalized with severe COVID-19-related pneumonia has failed in a late-stage trial.

The British drug maker, whose COVID-19 vaccine candidate is already among the most advanced, said the early-stage trial would evaluate if AZD7442, a combination of two monoclonal antibodies (mAbs), was safe and tolerable in up to 48 healthy participants between the ages of 18 and 55 years.

If the UK-based trial has a positive readout, AstraZeneca said it would proceed with larger, mid-to-late-stage trials to test AZD7442 as both a preventative treatment for the disease and a medicine for patients who have it, Reuters reported.

Monoclonal antibodies mimic natural antibodies generated in the body to fight off infection and can be synthesized in the laboratory to treat diseases in patients and has been endorsed by top scientists.

US based companies Regeneron and Eli Lilly are also testing mAbs-based treatments for COVID-19.

TEHRAN (Iran News) – Professor Ugur Sahin gave a speech on “Developing COVID-19 mRNA vaccine” during the 7th Science and Technology Exchange Program (STEP) on May 28.

The 2019 Mustafa(pbuh) Prize laureate, immunologist Ugur Sahin, talked about an mRNA-based vaccine that he and his research team have developed which was recently tested on human beings.

“We are facing a COVID-19 pandemic the only solution to which is a vaccine. This pandemic will challenge us for the next two years ”

Explaining the concept of mRNA vaccines, he said that “they deliver mRNA-encoded genetic information of the vaccine antigen to the cells in the body of the subject. It is particularly used in pandemics.”

“mRNA vaccines can be designed and manufactured fast within weeks; they have intrinsic immune-stimulatory properties and do not require the addition of adjuvants; they are highly immunogenic and induce neutralizing antibodies as well as T-cell responses. They are also well-characterized biopharmaceuticals with high purity and are free of animal material,” he added.

According to Sahin, the clinical trial protocol in Germany started April 23rd, 2020, with a target population of 200 healthy subjects aged 18 to 55. The objective of this was safety, tolerability, immunogenicity, in addition to determining the optimal dose for further studies.”

He also announced that the first results of this study are expected by the end of June/early July.

“We have started to increase our manufacturing. I’m confident this can be accomplished in the next three months and be distributed in a large dose. Depending on the dose of the vaccine, we can serve larger populations of people,” Sahin remarked.

He said: “We are trying to find a solution to allow free access of this vaccine to all with the cooperation of WHO.”

TEHRAN (Iran News) – Scientists have discovered an antibody which prevents the coronavirus from infecting human cells in “groundbreaking research” which could lead to the development of new treatments.

Building on research into the SARS coronavirus, scientists from Utrecht University in the Netherlands, as well as the Erasmus Medical Centre and the company Harbour BioMed (HBM), have identified a potential method of neutralising COVID-19.

They discovered that an antibody which prevents the SARS virus from infecting human cells could also block the novel coronavirus from infecting human cells too, according to a peer-reviewed study published on Monday in the journal Nature Communications, Sky News reported.

Testing their collection of antibodies on cultured human cells, researchers discovered one which binds to a specific part which is present in both SARS and the virus causing COVID-19.

The discovery could offer an initial step towards developing a fully-human antibody to treat or prevent the disease, which has infected more than 3.5 million people worldwide and led to more than 247,000 deaths.

The neutralizing antibody “has potential to alter the course of infection in the infected host, support virus clearance or protect an uninfected individual that is exposed to the virus,” said Dr Berend-Jan Bosch, co-lead author on the study.

Dr Frank Grosveld, the study’s other co-lead author, said the discovery provided “a strong foundation for additional research to characterise this antibody and begin development as a potential COVID-19 treatment”.

“The antibody used in this work is ‘fully human’, allowing development to proceed more rapidly and reducing the potential for immune-related side effects,” he added.

The fully-human antibody is different from conventional therapeutic antibodies, which are often first developed in other species before being “humanised” so they can be transmitted to people.

It was developed using Harbour BioMed’s H2L2 transgenic mouse technology – effectively a mouse which has been genetically engineered to contain human genes, enabling researchers to develop “human” antibodies without testing them on living people.

“This is groundbreaking research,” said Dr Jingsong Wang, the chief executive of HBM.
